一种2型糖尿病肾虚证唾液蛋白标记物的检测方法,其特征在于,具体步骤如下:(1)样品蛋白提取:样本中加入5%的裂解缓冲液进行涡旋混匀,超声60s,振幅22%室温提取30min;15000r/min、4℃离心20min,取出上清;收集上清,分装分装后冻存于‑80℃;(2)蛋白定量:采用Bradford法测定提取的蛋白浓度,先将样本用裂解缓冲液进行一定倍数稀释使其终浓度落在标曲范围内,稀释好的样本和标准品各取10μl分别和300μl蛋白定量染料避光反应20min,用酶标仪同时测定标准品和样本在595nm下的吸光值,根据标准品每管吸光值和浓度的关系绘制标准曲线,根据曲线公式计算各样品蛋白浓度;(3)蛋白酶解:蛋白定量后取200μg蛋白溶液置于离心管中;加入4μl还原剂,60℃反应1h;加入2μl的半胱氨酸阻断剂,室温反应10min;将还原烷基化后的蛋白溶液加入10K的超滤管中,12000r/min离心20min,弃掉收集管底部溶液;加入iTRAQ试剂盒中的溶液缓冲液100μl,12000转离心20min,弃掉收集管底部溶液,重复3次;更换新的收集管,在超滤管中加入胰蛋白酶,总量4μg,体积50μl,37℃反应过夜;次日,12000r/min离心20min,酶解消化后的肽段溶液离心于收集管底部;在超滤管中加入50μl溶解缓冲液,12000r/min再次离心20min,与上步合并,收集管底部共得到100μl酶解后的样品;(4)iTRAQ标记:从冰箱中取出iTRAQ试剂,平衡到室温,将iTRAQ试剂离心至管底;向每管iTRAQ试剂中加入150μl异丙醇,涡旋振荡,离心至管底;取50μl酶解后的样品转移到新的离心管中;将iTRAQ试剂添加到样品中,涡旋振荡,离心至管底,室温反应2h;加入100μl水终止反应;从4组样品中各取出1ul混合,用Ziptip脱盐后进行MALDI‑TOF‑TOF鉴定,确认标记反应良好;混合标记后的样品,涡旋振荡,离心至管底;真空冷冻离心干燥;抽干后的样品冷冻保存待用;(5)高pH条件下的反相色谱分离:所需试剂的调配:流动相A:98%ddH2O,2%乙腈,pH10;流动相B:98%乙腈,2%ddH2O,pH10;ddH2O用氨水调pH值到10;混合标记后的样品用100ul流动相A溶解,14000r/min离心20min,取上清待用;使用400μg酶解好的BSA进行分离,检测系统情况;取100ul准备好的样本上样;流速0.7ml/min,得到分离梯度数据;(6)ABI‑5600进行蛋白质分析:所需试剂的调配:流动相A:100%超纯水,0.1%甲酸;流动相B:100%乙腈,0.1%甲酸;将高pH反相分离得到的组份用20μl2%甲醇,0.1%甲酸复溶;12000r/min离心10min,吸取上清上样,上样体积10μl,采取夹心法上样;装载泵流速350nl/min,15min;分离流速350nl/min,得到分离梯度数据;(7)质谱数据分析:在选择数据库时,若为已经测序生物,直接选用该物种数据库,若为非测序生物,则选择与被测样品最为相关的大类蛋白质组数据库;采用ABI‑5600型质谱仪进行iTRAQ的质谱分析;(8)检测结果:对糖尿病肾虚证和正常对照组进行对比实验,定量蛋白质组学研究结果显示:鉴定1297个蛋白;与正常对照组相比,糖尿病肾虚证中鉴定到的1.5倍以上表达差异的显著差异蛋白共140个,其中上调蛋白64个,下调蛋白76个;差异蛋白列表如下:上调蛋白如下:1)检索号为Q07654,基因注释为Trefoil
