摘要: | 一种YN87448病毒毒株全基因组序列及其克隆方法,该克隆方法将全基因分1 5片段,用RT-PCR方法扩增相应cDNA,连入pGEM-T载体,利用通用引物测序,在病毒基因序旬扩增引物设计上,选择甲病毒保守区域的核苷酸序列设计引物,引物3’末端尽可能终止在简并密码子较少的氨基酸位置且最后一个碱基终止在氨基酸密码子的第二位碱基,本发明从分子水平阐明了本病毒与其他流行株的差异,对了解病毒的来源进化过程及分子流行病学有重要意义。也表明:它是预防辛德毕斯脑炎流行的最佳制备疫苗的候选株。 | ||
