摘要: | 本发明涉及以对活化血小板具有高亲和力的AnnexinV为载体构建血栓靶向性溶栓融合蛋白是AnnexinV与低分子量尿激酶融合基因在昆虫细胞中的活性表达产物。将AnnexinV与scu-PA-32K融合基因插入BactoBac昆虫杆状病毒表达系统的供体质粒中,将重组供体质粒转化钙细胞HB10B,并经蓝白斑筛选获取重组Bacmid,以脂质体法转染Sf21细胞而获得重组病毒BacUKAV。昆虫细胞Tn-5B1-4表达的AnnexinV-scu-PA-32K融合蛋白不仅对活化血小板具有高亲和力,而且具有尿激酶的纤溶活性,为双功能蛋白。 | ||
