本发明涉及一种细菌筛选和鉴定方法,属于生化鉴定技术领域。
背景技术
细菌筛选和鉴定应用最广泛、最基础的方法是生化鉴定方法。目前的细菌生化筛选和鉴定方法所涵盖的分类单元数量较少,一般仅有几十种,其中未囊括一些重要的细菌种类,如大肠埃希氏菌O157:H7、椰毒假单胞酵米面亚种(Burkholderia cocovenenans subsp.farinofermentans)等;而且,随着细菌名称的改变,有些筛选和鉴定方法对细菌的命名已经不够准确;此外,多数筛选和鉴定方法都需要先进行繁琐的染色、氧化酶等试验。
发明内容
针对上述筛选和鉴定方法存在的缺点,本发明的目的是提出一种更全面、更准确、更便捷的细菌筛选和鉴定方法。
本发明技术方案的步骤如下:
(1)确定细菌筛选和鉴定所需生化试验项目。选用常用的、方便配制的生化试验项目,舍弃操作繁琐的诸如硝酸盐还原、苯丙氨酸脱氨酶等常用试验。另外,考虑到鉴定时可以借用商业化生化鉴定条,因此尽量使用商业化生化鉴定系统中包括的生化项目。最终确定如下20项:β-半乳糖苷酶(ONPG)、精氨酸双水解酶(ADH)、赖氨酸脱羧酶(LDC)、鸟氨酸脱羧酶(ODC)、柠檬酸盐利用(CIT)、硫化氢产生(H2S)、尿素酶(URE)、吲哚(IND)、伏普(VP)、明胶液化(GEL)、甘露醇(MAN)、肌醇(INO)、山梨醇(SOR)、鼠李糖(RHA)、蔗糖(SAC)、蜜二糖(MEL)、苦杏仁苷(AMY)、阿拉伯糖(ARA)、乳糖(LAC)、木糖(XYL)。
(2)搜集各种菌株上述20项生化试验项目的阳性概率。通过查阅权威的鉴定手册和检测标准,如:《伯杰氏细菌鉴定手册》(第九版)、《伯杰氏细菌系统手册》(第二版)、《全国临床检验操作规程》(第三版)、《临床微生物学手册》(第七版)等,最终获得288个分类单元的20项生化试验结果,该288个分类单元如下:Acinetobacter baumannii/A.calcoaceticus,Acinetobacter lwoffi,Actinobacillus capsulatus,Actinobacillus equuli,Actinobacillus hominis,Actinobacillus lignieresii,Actinobacillus
