1.一种皮肤疾病基因SNP位点分型优化的方法,其特征在于,包括以下步骤:
1)提取样品中的基因组DNA,得到DNA提取液;
2)对步骤1)所得的DNA提取液进行PCR扩增,先加入DNA提取液与PCR缓冲溶液、Mg2+溶液、dNTP、DNA聚合酶和第一PCR引物混合反应,再加入等量的DNA提取液与PCR缓冲溶液、Mg2+溶液、dNTP、DNA聚合酶和第二PCR引物反应,得到PCR扩增产物;
其中,第一PCR引物由9个SNP基因位点组成,9个SNP基因位点在NCBI中SNP的数据库的rs号为:rs11966200、rs6010620、rs6902119、rs2393903、rs2431697、rs2236313、rs9318086、rs9468925和rs10490924;第二PCR引物由7个SNP基因位点组成,7个SNP基因位点在NCBI中SNP的数据库的rs号为:rs11200638、rs7007032、rs7331047、rs10088247、rs7325450、rs151823和rs1052134;
16个SNP基因位点的序列为:

3)PCR产物经核酸外切酶Ⅰ和虾碱性磷酸酶纯化,得到纯化后的PCR扩增产物;
4)纯化后的PCR扩增产物与延伸引物混合物、SNaPshot Multiplex Kit溶液和超纯水混合,进行延伸反应,得到延伸产物;其中,延伸引物混合物由16个SNP基因位点的引物组成,16个SNP基因位点在NCBI中SNP的数据库的rs号为:rs151823、rs2431697、rs7007032、rs10088247、rs2393903、rs6010620、rs11966200、rs9468925、rs2236313、rs6902119、rs9318086、rs7331047、rs7325450、rs10490924、rs11200638和rs1052134;16个SNP基因位点的序列为:

5)延伸产物经过虾碱性磷酸酶纯化,得到纯化后的延伸产物;
6)得到纯化后的延伸产物后,加入Hi-Di甲酰胺和Liz120 SIZE STANDARD混匀,变性后上测序仪;
7)通过测序仪分析步骤6)所得延伸产物的荧光标记和长度信息。
