1.一种固相多肽合成Exenatide的制备方法,其特征在于,包括如下步骤:
(1)制备Fmoc-Ser(tBu)-树脂
称取Rink Amide MBHA树脂100克,装入SHYM-25多肽合成仪中,用1000mlDMF浸泡30分钟,加入Fmoc-Ser(tBu)-OH 58g,HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干;
(2)制备Fmoc-Pro-Ser(tBu)-树脂
在步骤(1)的Fmoc-Ser(tBu)-树脂中,加入脱帽试剂800ml,25℃反应30分钟,抽干,用DMF、甲醇、DMF各洗涤数次,抽干,加入Fmoc-Pro-OH 52g、HBTU 58g、HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次抽干,获得Fmoc-Pro-Ser(tBu)-树脂;
(3)制备Fmoc-Pro-Pro-Ser(tBu)-树脂
在步骤(2)的Fmoc-Pro-Ser(tBu)-树脂中,加入脱帽试剂 800ml,25℃反应30分钟,抽干,用DMF洗涤,抽干,加入Fmoc-Pro-OH 52g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,获得Fmoc-Pro-Pro-Ser(tBu)-树脂;
Fmoc-Pro-OH的摩尔数为树脂的2~5倍;
(4)制备Fmoc-Pro-Pro-Pro-Ser(tBu)-树脂
在步骤(3)的Fmoc-Pro-Pro-Ser(tBu)-树脂中,加入800ml脱帽试剂,25℃反应30分钟,抽干,用DMF洗涤,抽干,加入Fmoc-Pro-OH 52g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解的混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,获得Fmoc-Pro-Pro-Pro-Ser(tBu)-树脂;
Fmoc-Pro-OH的摩尔数为树脂的2~5倍;
(5)制备Fmoc-Ala-Pro-Pro-Pro-Ser(tBu)-树脂
在步骤(4)的Fmoc-Pro-Pro-Pro-Ser(tBu)-树脂中,加入800ml脱帽试剂,反应,抽干,用DMF洗涤,抽干,加入Fmoc-Ala-OH 48g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,获得Fmoc-Ala-Pro-Pro-Pro-Ser(tBu)-树脂;
Fmoc-Ala-OH的摩尔数为树脂的2~5倍;
(6)制备Fmoc-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂
在步骤(5)的Fmoc-Ala-Pro-Pro-Pro-Ser(tBu)-树脂中,加入800ml脱帽试剂,反应,抽干,用DMF洗涤,抽干,加入Fmoc-Gly-OH 46g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,获得Fmoc-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂;
(7)制备Fmoc-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂
在步骤(6)的Fmoc-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂中,加入800ml脱帽试剂,反应,抽干,用DMF洗涤,抽干,加入Fmoc-Ser(tBu)-OH 58g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,抽干,获得Fmoc-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂;
(8)制备Fmoc-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂
在步骤(7)的Fmoc-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂中,加入800ml脱帽试剂,反应,抽干,用DMF洗涤,抽干,加入Fmoc-Ser(tBu)-OH 58g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,抽干,获得Fmoc-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro- Pro-Ser(tBu)-树脂;
(9)制备Fmoc-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂
在步骤(8)的Fmoc-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂中,加入800ml脱帽试剂,反应,抽干,用DMF洗涤,抽干,加入Fmoc-Pro-OH 52g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,获得Fmoc-Pro-Ser(tBu) -Ser(tBu)-Gly-Ala- Pro -Pro-Pro-Ser(tBu)-树脂;
(10)制备Fmoc-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂
在步骤(9)的Fmoc-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂中,加入800ml脱帽试剂,反应,抽干,用DMF洗涤,抽干,加入Fmoc-Gly-OH 46g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,获得Fmoc-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro- Pro-Ser(tBu)-树脂;
(11)制备Fmoc-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂
在步骤(10)的Fmoc-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly -Ala-Pro-Pro-Pro-Ser(tBu)-树脂中,加入800ml脱帽试剂,反应,抽干,用DMF洗涤,抽干,加入Fmoc-Gly-OH 46g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,获得Fmoc-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala -Pro-Pro -Pro-Ser (tBu)-树脂;
(12)制备Fmoc-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-Pro- Ser (tBu)-树脂
在步骤(11)的Fmoc-Gly-Gly-Pro-Ser(tBu) -Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂中,加入800ml脱帽试剂,反应,抽干,用DMF洗涤,抽干,加入Fmoc-Asn(Trt)-OH 90g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,获得Fmoc-Asn(Trt)-Gly-Gly -Pro -Ser(tBu)-Ser(tBu)-Gly-Ala-Pro- Pro-Pro-Ser(tBu)-树脂;
(13)制备Fmoc-Lys(Boc)-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro –Pro -Ser(tBu)-树脂
在步骤(12)的Fmoc-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-Pro- Ser(tBu)-树脂中,加入800ml脱帽试剂,反应,抽干,用DMF洗涤,抽干,加入Fmoc-Lys(Boc)-OH 72g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,获得Fmoc-Lys(Boc)-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂;
(14)制备Fmoc-Leu-Lys(Boc)-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro -Pro -Pro-Ser(tBu)-树脂
在步骤(13)的Fmoc-Lys(Boc)-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu) -Gly-Ala-Pro -Pro-Pro-Ser(tBu)-树脂中,加入800ml脱帽试剂,反应,抽干,用DMF洗涤,抽干,加入Fmoc-Leu-OH 54g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,获得Fmoc-Leu- Lys(Boc)-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro -Pro-Ser (tBu)-树脂;
(15)制备Fmoc-Trp(Boc)-Leu-Lys(Boc)-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly- Ala-Pro-Pro-Pro-Ser(tBu)-树脂
在步骤(14)的Fmoc-Leu-Lys(Boc)-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly -Ala -Pro-Pro -Pro-Ser(tBu)-树脂中,加入800ml脱帽试剂,反应,抽干,用DMF洗涤,抽干,加入Fmoc-Trp(Boc)-OH 64g、HBTU 58g,HOBT 21g,用1000ml接肽试剂溶解混合物,25℃反应1小时,抽干,分别用DMF、甲醇、DMF洗涤数次,抽干,获得Fmoc-Trp(Boc)-Leu-Lys(Boc)-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro -Pro - Ser(tBu)-树脂;
(16)制备Fmoc-Glu(OtBu)-Trp(Boc)-Leu-Lys(Boc)-Asn(Trt)-Gly-Gly-Pro-Ser (tBu) -Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂

在步骤(38)的Fmoc-Gly-Glu(OtBu)-Gly-Thr(tBu)-Phe-Thr(tBu)-Ser(tBu)-Asp(OtBu) -Leu-Ser(tBu)-Lys(Boc)-Gln(Trt)-Met-Glu(OtBu)-Glu(OtBu)-Glu(OtBu)-Ala-Val-Arg(Pbf)-Leu-Phe-Ile-Glu(OtBu)-Trp(Boc)-Leu-Lys(Boc)-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-树脂中,加入预冷至0℃的切肽试剂:TFA/EDT/H2O/TIS=1260ml/56ml/35ml/14ml,25℃反应3小时,减压除溶剂后,加入1500ml乙醚沉淀,收集沉淀物,用乙醚洗,在室温,置干燥器中,P2O5存在下减压干燥12小时,获得切肽后的Exenatide粗品,100g;
将Exenatide粗品溶于重量浓度为5%乙酸中,过滤,滤液经C18柱纯化,流动相:0.2M醋酸盐缓冲液∶乙腈为7.5∶2.5,体积比;流速为350ml/min;检测波长为:280nm;用液相色谱仪跟踪收集所需要的流出液,样品峰合并后去盐,冻干,获得产品12.2g,总收率为9.7%;
所说的接肽试剂为:NMM:DMF=1:5~15,体积比;
所说的脱帽试剂为:PIP:DMF=1:2~5,体积比。
