摘要: | 盖塔病毒全基因组序列、扩增方法及引物,该基因组全长包括11690个核苷酸,不包括5’端帽子结构、3’端的多聚核苷酸尾,整个基因组编码一个多聚蛋白,5’端2/3编码非结构蛋白,3’端1/3编码结构蛋白;扩增用引物覆盖整个病毒基因组,扩增片段长度为900-1000个碱基,相邻片段之间有70-120个碱基的重叠区,所有引物不包含简并碱基,100%与盖塔病毒序列相同,引物的3’端为C,G或T保证稳定地与cDNA序列结合,所有引物的退火温度,GC含量基本相同,从而保证所有引物能够在相同的PCR条件下进行扩增反应。本发明的盖塔病毒全基因组序列为制备盖塔病毒全基因组克隆,研究盖塔病毒结构与功能的关系、盖塔病毒与宿主相互关系及对盖塔病毒的预防与控制打下了基础。 | ||
